David Jon Gilmour nació en Cambridge, Reino Unido, hijo de un zoólogo profesor en la Universidad de Cambridge y de una profesora y editora de cine. Acudió a la escuela Perse en Cambridge, situada al lado del instituto en el que cursaba sus estudios Syd Barrett, de quien se hizo amigo. David y Syd solían juntarse a tocar la guitarra, y David le enseñaba a tocar canciones de los Rolling Stones. En 1963 formó la banda Joker's Wild, con algo de popularidad localmente, que abandonó en 1966 para viajar por España y Francia con unos amigos. La mala situación en la que vivía, llegando incluso a ser tratado en un hospital por malnutrición y tomar empleos casuales como modelo, hizo que regresase a Inglaterra en 1967.
Gilmour entró en Pink Floyd en enero de 1968, convirtiendo a la banda en un quinteto durante un corto período. En esos momentos, su trabajo consistía en sustituir a Syd Barrett cuando éste no podía continuar con los conciertos. Cuando Barrett dejó el grupo, Gilmour se convirtió en el guitarrista y en uno de los vocalistas (junto con Roger Waters y Richard Wright) del grupo. Después de la publicación de The Dark Side of the Moon y Wish You Were Here, Waters se hizo con el control de la banda, componiendo la mayoría de las canciones de Animals y The Wall. Wright fue despedido durante la grabación de The Wall, y las relaciones entre Waters y Gilmour se deterioraron durante las sesiones de The Final Cut y el rodaje de la película The Wall.
